Can the Adafruit Mega Proto Shield (product 192) be used on the Arduino Due?

Yes. The header layout is the same.

Will we be able to replicate both the reset and erase buttons?

The Reset pin is in the same location on both boards, so the reset button will work the same. I'm not sure what "erase" button you are referring to.
